Clarify requirements for autologous apoptotic cells to serve as antigen cargo for human dendritic cells triggering T-cell activation

Inadequate processing of phagocytosed antigens, apoptotic cells or pathogens can contribute to human disease, for example due to defects in the elicitation of adaptive immune responses. In the case of apoptotic cells, inadequate presentation of peptides derived from internalized apoptotic cells as peptide/MHC complexes may cause on the one hand autoimmunity (if self-antigens are presented to self-reactive T- or B-cells in an immunogenic manner) or on the other hand defective defense against tumor cells (if cancer-associated peptides are not presented effectively). This project aims to clarify in human dendritic cells mechanisms that determine which protein determinants are presented and trigger adaptive immunity, and which are fully degraded.
